Biological Activity
Description: Danshenol A has strong aldose reductase (AR) inhibitory activity, has IC50 of 0.10 microM which is comparable to that of epalrestat in clinical use. Danshenol A also has antitumor activity in T-24, QGY, K562, Me180 and BIU87 cell lines, shows inhibited growth of K562 (IC50 = 0.53 μg/mL), T-24 (IC50 = 7.94 μg/mL), QGY (IC50 = 4.65 μg/mL) and Me180 (IC50 = 6.89 μg/mL) cell lines.

Two new abietane diterpenoids from Salvia yunnanensis.[Pubmed: 16450305]

METHODS AND RESULTS:
Two new abietane diterpenoids, yunnannin A and danshenol C, were isolated from Salvia yunnanensis together with ten known diterpenoids, Danshenol A, przewalskin, tanshinone IIA, tanshinone I, crypotanshinone, 1,2-dihydrotanshinone, tanshinlactone, 5,6-dehydrosugiol, 12-hydroxy-6,7-seco-8,11,3-abietatriene-6,7-dial and phytol. Their structures were established based on spectroscopic data, chemical reactions and comparison with literature data.
CONCLUSIONS:
Compounds were tested for their antitumor activity in T-24, QGY, K562, Me180 and BIU87 cell lines. Compound showed inhibited growth of K562 (IC50=0.53 microg/mL), T-24 (IC50=7.94 microg/mL), QGY (IC50=4.65 microg/mL) and Me180 (IC50=6.89 microg/mL) cell lines while compound was inactive. Compound showed moderate inhibitory activity on QGY (IC50=16.75 microg/mL) and Me180 (IC50=5.84 microg/mL) cells.

Aldose reductase inhibitory constituents of the root of Salvia miltiorhiza Bunge.[Pubmed: 9377780]

METHODS AND RESULTS:
The constituents of the MeOH extract of Salvia miltiorhiza BUNGE, which showed strong aldose reductase (AR) inhibitory activity, were examined, and two new abietane-type diterpenoids, Danshenol A (1) and danshenol B (2), were isolated together with six known ones: dihydrotanshimme I (3), cryptotanshinone (4), tanshinone I (5), tanshinone IIA (6), (-)-danshexinkun A (7), and sugiol (8). Among them, 4, 5, and 8 were weak AR inhibitors with IC50 from 4.80 to > 10.0 microM, while 1, 2, 3, 6, and 7 were strong inhibitors (IC50 from 0.10 to 1.75 microM). Danshenol A (1), the strongest inhibitor, had IC50 of 0.10 microM which is comparable to that of epalrestat in clinical use.
CONCLUSIONS:
Moreover, from a consideration of IC50, and yield of each compound, it was concluded that tanshinone IIA (6) is the major active constituent of the MeOH extract and Danshenol A (I) and (-)danshexinkun A(7) are the minor ones.
